Transgenosis approach in the development of pea lines with improved resistance to viral pathogens. Proceedings from XVIIth Czech and Slovak Plant Protection Conference, 12-14 December, Prague, pp. 116-120, 2006.ISBN: 80-213-1564-4

  • Original language description

A set of PEMV and PSbMV isolates collected on pea plants in five locations in the Czech Republic (determined by symptoms on pea plants and confirmed by ELISA) was characterized by biological and molecular tools. The most severe isolates were selected forsequencing and the development of constructs for pea transformation. The strategy of induction resistance to viruses was based on pathogen-derived resistance (PDR) and post-transcriptional gene silencing (PTGS). Commercial cultivars of peas were transformed and T1 seed generations of GUS/PCR positive transgenic plants were obtained. Viral infection of the progeny of pea plants will be studied further.

  • Czech description

Na 5 lokalitách ČR by uskutečněn sběr izolátů virových patogenů PEMV a PSbMV (determinace symptomy na rostlinách a ELISA), které byly dále charakterizovány biologickými a molekulárními testy. Izoláty s nejvyšší patogenitou bly vybrány pro sekvenování a vývoj konstruktu pro transformaci hrachu. Strategie indukce rezistence byla založena na pathogen-derived resistance (PDR) a post-transkripčním urišování genů (PTGS). Komerční odrůdy byly transformovány a byly získány T1 rostliny s GUS/PCR pozitivní reakcí. Na těchto rostlinách bude dále studována reakce na virovou infekci.
