This study determined a role of Hsp70 in response of rabbit preimplantation embryos (Es) to hyperthermia (HT) in vitro. The exposure of Es to 41.5 oC either before or after DB did not affect blastocysts (Bl) rate, T-index and E cell number. Blocking of Hsp70 before DB resulted in no Es (0%) at Bl stage. From Hsp70-blocked Es after DB, 27.6% (ST+anti-Hsp) and 22.6% (ET+anti-Hsp) of Bl were formed, versus 71.4% (ST) and 69.7% (ET) in non-Hsp70-blocked Es. T-index of Hsp70-blocked Es before DB increased from 6.8% (ST) to 28.6% (ST+anti-Hsp) and from 6.1% (ET) to 29.6% (ET+anti-Hsp), whilst cell number was dropped from 111 +- 8.1 (ST) to 20.5 +- 2.42 (ST+anti-Hsp70) and from 106.8 +- 8.4 (ET) to 15.4 +- 1.34 (ET+anti-Hsp70). Oppositely, in Hsp70-blocked Esafter DB, neither TUNEL-index nor cell number were changed comparing to non-blocked Es.

Czech description

Cílem této studie bylo určit roli Hsp70 v odpovědi králičích preimplantačních embryí (Es) k hyperthermii (HT) in vitro. Vystavení Es teplotě 41.5 oC před nebo po diferenciaci blastocyst (DB) neovlivnilo podíl blastocyst (Bl), T-index a počet embryonálních buněk. Blokování Hsp70 před DB ukázalo, že Es (0%) se nevyvíjela do stádia Bl. Z počtu Es Hsp70-blokovaných po DB, se 27.6% (ST+anti-Hsp) a 22.6% (ET+anti-Hsp) formovalo do Bl, oproti Es Hsp70 neblokovaným to bylo 71.4% (ST) a 69.7% (ET).
