Esterase isozymes, proteins extracted in 2% glutathione buffer (separated in non denaturing PAGE system) and seed proteins separated in SDS page were used as biochemical markers for evaluation of polymorphysm level in three spelt wheat varieties, three breeders spelt lines (orig. from hybrids. between spelt and common wheat) and reference common wheat variety. Electrophoresis phenotypes and zymograms were evaluated by means of digital image analysis and Nei + Li coufficient if similarity was used to evaluate the relation of analysed genotypes. Entire evaluation of three marker systems showed differences between common wheat variety Brea and spelt varieties and spelt breeders´ lines. Also significant differences between old spelt varieties (Hercule, Altgold and Rouquin) and new spelt breeders´ lines were found.
