Effects of ions on the motility of fresh and demembranated paddlefish (Polyodon spathula) spermatozoa.
Original language description
Motility parameters of paddlefish (Polyodon spathula) spermatozoa, in different environmental conditions were investigated in this study. Paddlefish spermatozoa demonstrated the following characteristics: 100% of the cells were motile 10 s after activation with velocity 130-160 #m.s-1, then after 2 min velocity decreased to 80-130 #m.s-1; the period of motility was up to 9 minutes. Concentrations between 0.5 to 5 mM of KCl prevented activation of spermatozoa. When transferred into a swimming medium (20mM tris, pH 8.2 and 1 mg/ml BSA) with 0.5 mM KCl (combined 5 mM of NaCl or MgCl2) 80-100% cells were motile with velocity around 120-150 #m.s-1. MgCl2 significantly improved velocity of sperm motility at 10, 40, 50 and 60 s after activation with stable velocity of sperm around 140 #m.s-1. Very low concentration of 0.125 mM CaCl2 combined with 0.5 mM KCl initiated motility in 20% of sperm while all sperm were activated after 2 minutes with 0.25 mM CaCl2 in similar medium for the full peri
