    Determination of D- and L-amino acids produced by cyanobacteria using gas chromatography on Chirasil-Val after derivatization with pentafluoropropyl chloroformate

    A rapid and simple method was developed for the determination of free amino acids (AAs) released from cyanobacteria. The procedure involves trapping of AAs from the centrifuged cyanobacterial culture fluid on a cation-exchange resin, their release together with the resin by a direct treating with the reaction medium, followed by immediate derivatization with a corresponding chloroformate. The extractive alkylation does transfer the analytes into an organic phase, aliquot of which is subjected to GC analysis. Identification and quantification of AAs was performed by GC/MS and GC/FID, respectively, using propyl chloroformate (PCF) as the derivatization reagent. For chiral analysis, the cyanobacteria extracts were treated with 2,2,3,3,3-pentafluoropropylchloroformate (PFPCF) to create more volatile analytes. Separation of the AA enantiomers was accomplished on a Chirasil-Val capillary column and the D/L- enantiomeric ratios were determined.

    Byla vyvinuta rychlá a jednoduchá metoda stanovení volných aminokyselin uvolňovaných ze sinic. Postup zahrnuje zachycení aminokyselin z centrifugátu kultury sinic na katexu, jejich uvolnění spolu s iontoměničem přímým působením reakčního media a okamžitou derivatizaci příslušným chlormravenčanem. Analyty jsou přeneseny do organické faze extraktivní alkylací. Identifikace a kvantifikace aminokyselin byla prováděna GC/MS a GC/FID, s využitím propylchlormravenčanu jako derivatizačního činidla.Pro chirálníanalýzy byly připraveny těkavější deriváty reakcí s 2,2,3,3,3-pentafluorpropylchlormravenčanem. Separace enantiomerů byla provedena na kapilární koloně Chirasil-Val a byly zjištěny podíly D/L-aminokyselin v kulturách sinic. Aminokyseliny uvolňované kulturami sinic mohou ovlivňovat tok energie a nutriční hodnotu sinic ve vodním potravním řetězci a take mezibuněčnou komunikaci v kulturách sinic.
