Stable Agrobacterium-mediated transformation of Norway spruce embryogenic tissues using somatic embryo explants

  • Original language description

In conifers and other plants with long reproductive cycles, transformed embryogenic calli can serve as a convenient source of plant material for the testing insecticidal or fungicidal transgene efficiency. In this report, transgenic embryogenic callus was obtained after transformation of somatic embryos of Norway spruce (Picea abies) by Agrobacterium tumefaciens with the gus-intron chimeric gene. The stable integration of transgenes was confirmed by PCR and Southern hybridization. The transformation wassuccessful only with suitable embryogenic line sensitive to Agrobacterium. From nine embryogenic lines tested only one gave transgenic callus
