Analysis of recombinant anticancerogenic nuclease RTBN1 in ?leaf factory? system upon co-expression of modified genes
Original language description
RTBN1, which is a nuclease with antitumor activity, is currently produced for biomedical experiments using ?leaf factory system. To study stability of its expression, several potential plant expression modificators were cloned and co-expressed with nuclease using A. tumefaciens infiltration system. The nuclease analyses revealed that nuclease TBN1 is quite stable in the used ?leaf factory system as to its molecular mass and glycosylation pattern, only some quantitative changes of nuclease activity weredetected. Our experiments showed that activity and production of the nucleases increased significantly upon co-infiltration with silencing suppressor p19 and decreased dramatically upon co-expression with morphogenesis changing transcription factor HlMyb3. Selected glycosyltransferases and kinases influenced activity and production only insignificantly.
