Original language description
Cells and multicellular organisms share the ability to activate an internal death program known as apoptosis. Apoptosis theory, developed over the last 20 years, has challenged conceptual thinking in all aspects of cell biology. In cancer, the increase in the tumor burden can be regarded as the result of too much growth and too little death of the tumor cells. Therefore, the aim of chemotherapy and radiotherapy is to block proliferation and to enhance cell death. One of the major signaling pathways initiated by DNA damage and other cytotoxic stresses involves the induction of TP53 protein. Via induction of TP53, DNA damage can induce growth arrest or programmed cell death in many cell types. This induction response appears to be a major determinant ofresponses to chemotherapy in certain tumor types, such as those derived from lymphoid cells or germ cells.
