Reactivation of AChE and BuChE activity - possible implications for prophylaxis against nerve agents
Original language description
The primary targets of OP, such as pesticides (paraoxon, malathion, chlorpyrifos) and nerve agents (sarin, soman, tabun, VX) are enzymes from the group of cholinesterases - AChE; EC 3.1.1.7 and; EC 3.1.1.8. Antidotes currently used for OP intoxications consist of a combination of prophylaxis with carbamates (mostly pyridostigmine) and therapy with anticholinergics (atropine), AChE oxime reactivators (pralidoxime, oxime HI-6), anticonvulsant drugs (diazepam). Another approach for prophylaxis is administration of suitable scavenger, which is capable of neutralizing OP rapidly after its penetration into organism. For these purposes, catalytic (paraoxonase, phosphotriesterase) and stoichiometric (cholinesterases) enzyme scavengers are intensively investigated. According to our results, reactivability of inhibited AChE is almost two-times higher, than that of BuChE. For construction of effective pseudocatalytic scavenger, more potent compounds are needed.
