  • Original language description

    The present study describes the determination of mycotoxins deoxynivalenol (DON) and zearalenone (ZEA) in barley from Brazil and in malted barley. The quality of the raw materials involved in the malting process plays a decisive role in the creation of a safe beer product. Immunoaffinity columns for extraction and clean-up, and liquid chromatography coupled with mass spectrometry (LC-MS) are the most commonly used method for determining the presence of mycotoxins. A total of 24 samples were analysed for DON and ZEA. The average concentration of DON and ZEA was 2859.5 μg/kg and 727.9 μg/kg for barley and 1057.3 μg/kg and 275.1 μg/kg for malt, respectively. The content of mycotoxin decreases by about 63% during the malting process. The validation parameters of an analytical method such as linearity, limit of detection (LOD) and limit of quantification (LOQ), and recovery were tested. The calibration curves were linear at least in the range of 10–1000 ng/mL, LOD and LOQ ranging 2.8–3.5 μg/kg and 9.2–11.6 μg/kg, respectively. Recovery for DON and ZEA ranged 87.6%–92.9% with a RSD range of 2.9%–9.2% and 89.6%–105.7% with a RSD range of 0.4%–5.3%, respectively.
