    High incidence of Peach latent mosaic viroid (PLMVd) and Hop stunt viroid (HSVd) infections of peach trees was recently found in the Czech Republic. In the frame of prepared certification system for fruit trees planting material a PCR method for these purposes had to be developed. Several primer pairs were designed according to the published sequences of both viroids. Various plant material (leaves, flowers, pollen, bark) was tested during the year and also several methods of RNA extraction were tested.PCR with different primer pairs gives reliable results. More important is the time of testing, material, and RNA extraction method. The best period for detection of both viroids is late spring and early summer taking leaves as testing material and usingany of RNA extraction methods. For older leaves in late summer using of Qiagen kit for RNA extraction can be recommended. Detection of both viroids in bark is possible even during winter but it is not very reliable.
