Ploidy level assessment of the Czech kochia (Kochia scoparia (L.) Schrad.) biotypes resistant to acetolactate synthase inhibitors and atrazine

  • Original language description

Kochia biotypes (Kochia scoparia (L.) Schrad.) resistant to acetolactate synthase (ALS) inhibitors and atrazine were proved at railway junctions in the Czech Republic. Susceptible biotypes were found in several localities in the proximity of railway stations, roads and parking areas, and on arable land with common crop rotation and herbicide use. Ploidy levels of both susceptible and resistant biotypes of kochia were investigated by flow cytometry of vegetative tissues. All examined plants belonged to DNA-diploids, nevertheless, endopolyploidy was detected in all vegetative parts. In leaf tissues, nuclei with 2C DNA content were usually the most abundant group (mean proportion 55.2 %), followed by 4C nuclei (37.6 %) and 8C nuclei (7.2 %). No significant difference in endopolyploidy profile between susceptible and resistant biotypes was observed. Generally, older tissues showed higher level of endopolyploid nuclei indicating certain age-related correlation. Nuclei isolated from stems an
