    The seeds of Nigella sativa L. (Ranunculaceae) known also as black cummin are widely used in traditional medicine in the Middle East and South Asia for the treatment of various respiratory and gastrointestinal diseases, pain in joints and eczema. Recentstudies have demonstrated its anticancer, anti-inflammatory, antihistaminic, antibacterial and analgesic activities. Many of these activities are attributed to quinone constituents, of which thymoquinone is more abundant compound. We report here resultsof an in vitro inhibition of cyclooxygenase-1 (COX-1) and ?2 (COX-2) activity for by thymoquinone, the main quinone compound of N. sativa seeds. The radiochemical assay was based on the inhibition of prostaglandin E2 and D2 biosynthesis from [14C] radioactive arachidonic acid catalyzed by COX-1 and ?2 respectively. The identification and quantification of the COX reaction metabolites were analyzed by HPLC performed on C18 reversed phase column with an on-line radioactivity flow detector.
