    The impact of the altered polyprotein ratios on the capsid assembly and infectivity of Mason-Pfizer monkey virus

    Mason-Pfizer Monkey Virus (M-PMV) belongs to the Betaretrovirus genus within the family Retroviridae and causes immunosuppressive disease of Rhesus monkey. M-PMV genome consists of four genes in the following order: 5' gag-pro-pol-env 3'. Retroviruses regulate the ratios of the primary translation products, polyprotein precursors Pr78 (Gag), Pr95 (Gag-Pro) and Pr180 (Gag-Pro-Pol), by frameshifts during translation of the individual genes in a host cell. Two frameshifts, first one at the end of gag geneand the second one at the end of pro gene, occur approximately with the efficiency of 10-20%. M-PMV structural proteins are present at the tenfold higher level than the viral enzymes. The ratio of polyprotein precursors varies in the individual retroviruses and represents a regulatory mechanism controlling relative amounts of protease and its substrates.
