Retroviruses synthesize their structural proteins and enzymes as polyprotein precursors. The number and composition of these polyprotein precursors varies and represent the control mechanism of the structural proteins and enzymes ratio. Mason-Pfizer monkey virus (M-PMV) synthesizes three polyprotein precursors, Pr78 (Gag), Pr95 (Gag-Pro), Pr180 (Gag-Pro-Pol), using two ribosomal frame-shifting events occurring with the frequency of 5 - 20% each. We mutated M-PMV frame-shift sites in order to mimic the ratio of polyprotein precursors in HIV and RSV, so that the mutants produce only Gag and Gag-Pro-Pol (HIV-like mutant) or Gag-Pro and Gag-Pro-Pol (RSV-like mutant) polyproteins, respectively. We investigated the effect of such mutations on the ability toassemble the proper particles, their maturation and consequently the virion infectivity.
