Retroviruses regulate the ratios of their polyprotein precursors by frame-shifting events during translation of the genome-length DNA in the host cell. Mason-Pfizer Monkey virus (M-PMV) uses two ribosomal frameshifts in order to synthesize its polyprotein precursors, Pr78 (Gag), Pr95 (Gag-Pro), Pr 180(Gag-Pro-Pol). The nature of polyprotein precursors varies in retroviruses. In order to investigate the requirement for specific polyprotein precursors in assembly, maturation and consequently the virion infectivity, we mutated frame-shifting sites in the M-PMV genome. The resulting viral mutants mimic the polyprotein ratios of HIV or RSV producing only Gag and Gag-Pro-Pol or Gag-Pro and Gag-Pro-Pol polyproteins, respectively. The mutant producing only Gag-Pro-Pol or Gag and Gag-Pro polyproteins were also generated. Properties of these mutants were compared to wild type virus and D26N mutant with inactive proteaseSimultaneous abrogation of both frameshift sites, which results in synthesis
