The target of this work is to increase biodegradation of polychlorinated biphenyls (PCBs) by the plants transformed with the gene from bacterial PCB degradation pathway. Bacterial aerobic PCB degradation pathway contains 4 steps (bph operon) in which thethird one (bphC) catalyzes the conversion of 2,3-dihydroxybiphenyl into 2-hydroxy-6-oxo-6-phenylhexa-2,4-dienoic acid and thus destroyes the biphenyl ring. For this purpose the bphC gene encoding the enzyme 2,3-dihydroxybiphenyl-1,2-dioxygenase was chosen from bacteria Pseudomonas testosteroni B-356 for cloning into plant Nicotiana tabacum. Different constructs in cassette with bphC containing genes for GFP (green fluorescent protein), GUS (glucuronidase) and LUC (luciferase) were prepared to allow sensitive detection of bphC expression in plants.
