Fractionation of Co, Cu, Fe, Mn, Mo, Ni, P and Zn species in extracts of buckwheat semolina and amaranth flour in 0.02 moll-1 Tris-HCl (pH=7.5) buffer was performed by SEC/ICP-MS hyphenation. Element specific chromatograms of the buckwheat extract are similar to those of legumes and cereals. Except for Fe and Mn the remaining metals are concentrated in a low-molecular fraction (Mr<2 kDa). Fe is bound in a high-molecular fraction (Mr>150 kDa). Chromatograms of Co, Fe, Mo and Ni species in amaranth extracts correspond with this pattern too. However, Cu is also bound in a medium-molecular fraction (Mr~40 kDa) and in a high-molecular one (Mr>150 kDa). The main part of Mn in both extracts is present in labile complex and ionic form. Fractions rich of tracemetals were isolated using preparative-scale SEC and the organic ligands of metals were consequently preconcentrated on Chelex 100 in Cu2+ form and eluted by ammonia solution.
