The acetone-butanol-ethanol (ABE) fermentation is a biotechnological process of solvent production by bacteria of the genus Clostridium. The aim of the work was to develop the method for viability determination of Clostridium pasteurianum and apply it for monitoring of ABE fermentation. First, seven fluorescent probes were tested in order to distinguish live and dead clostridial cells in suspension (propidium iodide, sytox green, ethidium bromide, fluorescein diacetate, 5(6)-carboxyfluorescein diacetate, rhodamine 123, bis-((1, 3-dibutylbarburic acid) trimethine) oxonol-BOX). BOX was chosen as the most convenient one for viability determination of C. pasteurianum and the method of staining was subsequently optimised (concentration of BOX: 1 ?g.ml-1, incubation time: 7 min, concentration of cells: OD600nm = 0.20 ? 0.01, demineralised water). Then the fluorescent staining method was applied to batch fermentation process carried out in a laboratory bioreactor under anaerobic conditions. F
