    Fractionation of herbal extracts using preparative high-performance liquid chromatography

    Many herbal extracts are known to contain biologically active molecules, some of them find uses in pharmaceuticals production. In the recent years, substances with a potential to inhibit lipolysis are being searched. One of potential applications is a treatment of some chronic diseases such as obstructive sleep apnea causing hypoxia, which results in adipocyte metabolism disruption. The aim of our study was to isolate and identify novel bioactive secondary metabolites from Artemisia scoparia, Panax ginseng and Phellodendron sp. plants that are presumed to possess a potential to reduce the extent of lipolysis. To obtain an entire set of compounds differing in their polarities, three extracts were prepared from each dried plant: aqueous, methanolic and dichloromethane. These extracts were then fractionated using preparative high-performance liquid chromatography Waters AutoPurification System coupled to single quadrupole mass spectrometry detector QDa. At the first phase, extracts separation was optimized using analytical column XBridge® C18 5 μm (4.6 x 100 mm). After scale-up to preparative mode employing reversed phase column XBridge® C18 5 μm (19 x 100 mm), time-defined fractions were collected. Obtained fractions were concentrated to measure antilipolytic activity. Enzymatic assay based on measurement of released free glycerol concentration was performed both for spontaneous and by isoprenaline induced lipolysis. Fractions that showed antilipolytic activity were further fractionated for more detailed investigation of their composition by ultra high-performance liquid chromatography coupled to high-resolution mass spectrometry with tandem Quadrupole Time-of-Flight mass analyzer. In this way, compounds possibly inhibiting lipolysis were searched.
