    GC–MS strategy for a comprehensive assessment of cannabis metabolome

    Cannabis is a very complex plant that provides several potential health benefits. However, these benefits strongly depend on the composition of the present compounds. In this study, we tested and critically evaluated two different sample preparation strategies (with/without derivatization employing N-Methyl-N-(trimethylsilyl)trifluoroacetamide, MSTFA, as a silylating agent) followed by gas chromatography coupled to mass spectrometry (GC–MS). Subsequently, we applied these methods to investigate the metabolome of two chemotypes, including five different varieties of Cannabis. The chemometric methods, such as principal component analysis (PCA) and partial least squares discriminative analysis (PLS-DA), were employed to provide the possibility of distinguishing cannabis varieties based on their metabolome. In the next step, significant compounds (‘markers’) for each variety (or varieties) were found and identified. For the identification of ‘markers’, the NIST library, in combination with an ‘in house’ developed database containing high-resolution mass spectra of 26 neutral cannabinoids and 38 MSTFA derivates, was used. © 2025
