    Detection of bioaffinity complexes in microchannels by fluorescence techniques.

    This work deals with multichannel microfluidic systems intended for bioapplications, particularly for medical diagnostics. Microfluidic systems consist of several parallel channels with small characteristic dimensions (10-100 ?m). These devices are fabricated from polystyrene (PS) and polydimethylsiloxane (PDMS) by a combination of microfabrication techniques ? UV photolithography, micromilling, thermal bonding, galvanic metallization, casting etc. In the experiments, the biological receptor is immobilized on the microchannel walls by a passive sorption. The blocking reagent 1 % BSA is used to cover empty regions on the walls. Then, aqueous solutions of ligands and detection reagents with a fluorescent marker are subsequently introduced in the channels. Finally, a solution of a detergent is used to break nonspecific bonds among the biomolecules.
