Original language description
We successfully proved the creation of spheroids by human colorectal adenocarcinoma cell line (HT-29) by usage of the liquid marbles, liquid droplets encapsulated by hydrophobic powder. During the cultivation cells spontaneously formed the spherical agglomerates (spheroids) without the scaffold. The study focused on the influence of different parameters (volume, cell density, day) on final harvest. To evaluate the harvest we counted the number of created spheroids and the total number of cells. Better simulation of living body is essential to increase the efficiency of drug development. In vivo conditions are significantly different comparing to in vitro conditions. Today used cell monolayers (2D structures of cells) are not accurate enough to simulate living organism, but growing cells as 3D structure have more potential. Drug absorption, distribution, metabolism, excretion and toxicity could be tested on these 3D multicellular spheroids, decrease the use of animal models and accelerate the drug development.
