Original language description
Intracellular compartmentalization is essential for cellular function, however, selectively targeting specific organelles remains a significant challenge in chemical biology. We report the development of smallmolecule probes that accumulate in specific organelles through the application of physicochemical tuning and targeting motifs. These probes incorporate bioorthogonal functional groups, enabling rapid and selective labeling reactions within living cells. Using tetrazine ligation and fluorescence imaging, we demonstrate organelle-specific visualization with high spatial resolution and minimal background. This platform opens new avenues for dissecting subcellular processes and engineering targeted chemical interventions. The developed method enables labeling and visualization of specific small molecules inside the organelles of living cells, which can be used in bioorthogonal reactions and for bioimaging.
