    The Fission Yeast Ortholog of eIF3a Subunit Is Not Functional in Saccharomyces cerevisiae

    The largest subunit of the translation initiation factor 3 eIF3a is an evolutionary conserved protein. Here we describe isolation of the eIF3a from Schizosaccharomyces pombe (SpeIF3a) and subsequent testing of its function in budding yeast Saccharomycescerevisiae. We found that formation of the endogenous eIF3 complex was not affected by the overproduction of SpeIF3a in S. cerevisiae but SpeIF3a was more tightly bound to S. cerevisiae ribosomes than the endogenous eIF3a. Its expression resulted in alteration of cell morphology

    Největší podjednotka translačního iniciačního faktoru 3 eIF3a je vývojově velmi konzervativním proteinem. V práci popisujeme izolaci sekvence kódující eIF3a v S.pombe (SpeIF3a) a testování funkce SpeIF3a v pučící kvasince S.cerevisiae. Zjistili jsme, žeani nadprodukce tohoto ortologu neovlivňuje tvorbu endogenního eIF3 komplexu, nicméně SpeIF3a je vázán na ribosomy pevněji než endogenní eIF3a. Jeho nadprodukce způsobuje výrazné defekty v morfologii buněk
