MCP-1-2518 (A/G) single nucleotide polymorphism in Czech patients with pulmonary sarcoidosis

  • Original language description

Background and aim: Genetic factors controlling production of chemotactic cytokines are thought to be important for development of inflammatory response in pulmonary sarcoidosis. Monocyte chemoattractant protein (MCP)-1/CCL2, a chemokine recruiting mononuclear cells, has been shown to play important role in sarcoidosis. A single nucleotide polymorphism of the MCP-1 gene promotor (-2518*A/G) has been associated with increased production of MCP-1 protein. This study, therefore, aimed at exploring relationship between this polymorphism and pulmonary sarcoidosis including Löfgren's syndrome (LS). Methods: The MCP-1-2518 genotypes were determined using polymerase chain reaction with sequence specific primers in 130 patients with sarcoidosis and 359 healthycontrol subjects. ELISA was used to detect serum (n=91) and BALF (n=57) MCP-1 protein level. Results: While no significant difference in the allele distribution between patients with sarcoidosis and healthy controls was observed (P=0.17),

Vztah mezi MCP-1-2518 (A/G) a sarkoidózou u pacientů českého původu

  • Czech description

Frekvence alely MCP-1-2518*G (p=0.01, relativní riziko, OR=2.3) a genotypu GG (p=0.03, OR=3.9) byla signifikantně zvýšená u pacientů s Löfgrenovým syndromem. Hladina proteinu MCP-1 v séru a BALF nekorelovala s MCP-1-2518 genotypem.
