The sensitive method for quantitative analysis of natural isoprenoid cytokinins (base, riboside, 9-glucoside, O-glucoside and ribotide) was developed. The method for estimation of 37 naturally occurring cytokinins is based on a baseline chromatographic separation of 20 cytokinins (trans/cis-zeatin, dihydrozeatin, isopentenyladenine, benzyladenine, meta/ortho-topoline groups). Ethanolic extracts containing a mixture of deuterium or 15N labeled standards were purified using a combined DEAE-Sephadex, octadecysilica column and immunoaffinity column based on wide-range specific monoclonal antibodies and analyzed by HPLC linked to a micromass single quadrupole mass spectrometer with an electrospray interface (HPLC/ESI-MS) and a photodiode array detector.
