Original language description
New approach in the analytical technologies of plant hormone analysis was introduced in the field of cytokinins and might be also soon applied in the analysis of other plant hormone groups. A new sensitive method for the quantitative analysis of all natural isoprenoid and aromatic cytokinins in plants is based on combined "generic" immunoaffinity chromatography - electrospray single-quadrupole mass. A baseline chromatographic separation of 20 non-derivatized naturally occurring cytokinins has been obtained. Precise analysis of O-glucoside and nucleotide fractions were also performed by the HPLC-MS runs but after an enzymatic treatment. More than 50 different cytokinin metabolites can be measured by this approach. When low cone voltage (20 eV) was applied, all studied cytokinins were determined in aqueous methanol as dominant quasi-molecular ions of M+H + with limits of detection from 10 to 50 fmol.
