    Production of reactive oxygen species by Photosystem II.

    Molecular oxygen released as a by-product of photosynthetic water oxidation acts in two different ways in the photosynthetic processes: 1) O2 is used as a sink for electrons and 2) O2 is a source of reactive oxygen species (ROS). Oxygen is reduced in successive univalent reactions to form superoxide radical (O2 -), hydrogen peroxide (H2O2) and hydroxyl radical (OH ). Production of O2 - was demonstrated using either an assay involving of cytochrome c reduction in the presence of xanthine/xanthine oxidaseor voltametric methods. The production of H2O2 was suggested to originate either from the dismutation of O2 - or incomplete oxidation of water. Using spin-trapping EPR technique it has been demonstrated that illumination of PSII membranes results in theformation of OH . In the present study, EPR spin-trapping spectroscopy was used to investigate light-induced production of ROS in PSII membranes.     Pomocí EPR spektroskopie byla studována tvorba reaktivních form kyslíku na PSII membránnách. Bylo zjištěno, že na akceptorové straně PSII dochází k tvorbě OH pomocí dvou možných mechanismů, které vedou k tvorbě vázaného a volného peroxidu vodíku. Získanévýsledky byly diskutovány vzhledem ke známým faktům.
