Globular and bipolar structures induced on the surface of tobacco leaves on a MS medium supplemented with 0.8 mg.l-1 2,4-D, 0.5 mg.l-1 BAP and 13.2 mg.l-1 aspartic acid formed callus and/or plantlets after their transfer on a MS medium without growth regulators. The content of plant growth regulators was studied in relation to growth of the callus and morphogenesis of plants from excised globules. The highest content of 2,4-D (2,4-dichlorophenoxyacetic acid) was detected in globular and bipolar structures isolated from leaf surface several weeks after their formation, similarly as callus on cultured leaves. Callus formation from these globules was accompanied by the decrease of 2,4-D content. A pronounced decrease of 2,4-D content was observed in the case of plantlet regeneration.
