Original language description
Direct organogenesis or callus formation of tobacco leaves was induced on a medium containing 0.8 mg.l-1 2,4-D, 0.5 mg.l-1 BAP and 13.2 mg.l-1 aspartic acid. Shoots or callus formation with globular structures depended on accumulation of 2,4-D in leavesand ethylene production. In long-term culture on medium with growth regulators the content of 2,4-D in leaves gradually increased, the highest 2,4-D content, similarly as the highest production of ethylene being detected after 2 weeks of culture. Globules excised from leaves emerged to shoots in culture where ethylene levels decreased by aeration, in contrary to culture without aeration in which callus was formed after excision. Short term culture (20 hours) of leaves on induction medium and subsequentculture on a basal medium resulted in direct organogenesis.
