2,4-D uptake and somatic embryogenesis Cucumber explants (Cucumis sativus L.)
Original language description
Cucumber hypocotyl explants formed the embryogenic suspension after three weeks of cultivation. There was a frequent occurrence of embryos but they showed developmental abnormalities combined with a low ability of plant regeneration. Debeaujon and Branchard (1993) noticed that the squash embryo abnormalities were related to the 2,4-D concentration in the induction medium. Embryogenic callus on the cut edges of cotyledons was formed after one week of cultivation. The occurrence of somatic embryos from the cotyledon explants was lower than those from the hypocotyl explants but some of the embryos proliferated and grew into flowering plants. The 2,4-D level in cotyledons increased faster than that in hypocotyls; the level of the 2,4-D in cotyledons already reached its maximum after 6 h but in hypocotyls after 32 h.
