Effect of stress factors on Norway spruce embryogenic culture induction
Original language description
Somatic embryogenesis could be result of plant survival strategy under stress conditions endangering their normal way of reproduction. Besides cold and oxygen deficit, used as pre-treatment, 2,4-D is one of the main stress factors inducing in vitro somatic embryogenesis on Norway spruce zygotic embryo. The injury of embryo is other stress factor, which could combine with 2,4-D and affect frequency of achieved polyembryogenic cultures. In this work the effect of longitudinal bisection of zygotic embryo was studied with regard to uptake and metabolism of 14C-2,4-D. Injury improved its influx and efflux and affected also its metabolic activity. The ratio between the content of free 14C-2,4-D and its polar and non-polar metabolites was higher in bisected zygotic embryos than in intact ones. Injury also increased the number of achieved embryogenic cultures.
