    The uptake of 14C-2,4-D To zygotic embryos of Norway spruce and induction of polyembryogenic cultures

  • Original language description

    The growth regulator 2,4-dichlorophenoxy acetic acid (2,4-D) is an important tool used in induction of somatic embryogenesis in most conifers. That is why uptake of 14C-2,4-D and induction of embryogenic cultures in intact zygotic embryos, in longitudinally halved and horizontally divided zygotic embryos of Norway spruce were compared. Results showed the highest 14C-2,4-D uptake per 1mg of fresh weight in intact radicles in the first 8 hours on induction medium and also the highest uptake in separated radicles after 4 hours in culture. Nearly no embryogenic cultures developed on this part. On the contrary the uptake of 2,4-D into cotyledonary part of embryos was the lowest and it was aligned with the highest number of achieved embryogenic cultures. Hypocotyls showed intermediate results in both parameters.
