Successful conversion of mature somatic embryos depends on balanced growth and development of both shoot and root. But the cotyledons and hypocotyl develop first and after that it can be difficult to get the root to grow in normal way. In our experimentswe used mature somatic embryos of two Norway spruce cell lines. The root growth of plantlets was stimulated by addition of indolyl-3-butyric acid (IBA) into conversion media. The supplement of 0.1 ?M IBA into the first conversion (germination) medium with activated charcoal was the most beneficial and increased root length approximately about 54-86% after 4 weeks. The plantlets were then transferred onto the second conversion medium with lower concentration of IBA, which was 0.05 ?M. The roots continued in growth and shoots were also well developed. After two months the plantlets were successfully transplanted into the soil.
