Morphological assessment of the reticulocyte maturation in anemic dogs using two different staining methods and the Sysmex XT-2000IV analyzer.

  • Original language description

Objective: The maturation of reticulocytes is defined by decrease of ribosomal RNA. However its identification using supravital stainings implements important result variability. Parallel analysis on validated Sysmex system recognizing reticulocyte fractions according to the fluorescence intensity may provide more detailed morphological evaluation of erythropoietic activity. Design and Method: The aim of this preliminary study was to assess reticulocyte maturation based on blood smear evaluation using new methylene blue (NMB) and brilliant-cresyl blue (BCB) in 15 anemic dogs. Sysmex XT 2000iV analyzer with subclassification of reticulocytes in low- (LFR), medium- (MFR) and high (HFR) fluorescence reticulocyte fraction was used as a reference method. Reticulocytes in all smears were subdivided in six groups: P1-P3 with punctate pattern (individual distribution or with linear arrangement) and A1-A3 if aggregations were noticed (corresponding to RNA content relative to reticulocyte surfac
