Direct ELLA: qualitative method for insects determination

The polysaccharide chitin and its subunits are found in large quantities in insects and participate in the construction of the entire exoskeleton. One of the ways to verify the presence of insects in food and thereby ensure their safety is to detect this polysaccharide, more precisely N-acetyl-D-glucosamine. Only lectin has the ability to bind specific polysaccharides, therefore this work was focused on optimizing the ELLA (enzyme-linked-lectin assay) method. Chitosan and an extract of the house cricket (Acheta Domesticus) were used as a model sample. The limit of detection (LODλ= 0.3532) was determined for this method. Chitosan samples at concentrations of 0.01 and 0.005 g/ml and cricket extract samples were confirmed as positive by the ELLA method.
