Gabapentin-lactam (GBP-L), a transformation product of the widely used anticonvulsant gabapentin (GBP), is formed by intramolecular cyclisation. It is often detected in aquatic environments due to its high chemical stability and poor biodegradability. Although the parent compound has been thoroughly studied, the ecotoxicological effects of GBP-L are not well understood, particularly at environmentally relevant concentrations. This study investigates the effects of GBP-L on the behaviour of zebrafish (Danio rerio) larvae following exposure to two concentrations: 1.5 μg/L and 100 μg/L. The results revealed a notable increase in total swimming distance in both groups compared to the control group. Both groups exposed to GBP-L exhibited increased activity during both the light and dark phases, indicating a disruption to normal behavioural patterns. While these results are consistent with previous findings for gabapentin, the reversed concentration-response pattern suggests differences in pharmacodynamics or toxicity thresholds between the two compounds. To our knowledge, this is the first study to demonstrate behavioural changes in zebrafish due to exposure to GBP-L. These findings highlight the importance of including pharmaceutical transformation products in environmental risk assessments, and further research is needed to understand their mechanisms of action
