Purification of Leptin Receptor from Human Serum

  • Original language description

Leptin plays a significant role in the body weight regulation and it is frequently used as a precise indicator and modulator of adipose tissue volume. Based on the general pharmacology.... The soluble part of leptin receptor (OB-Re), which is anchored tothe membrane, is an antagonist of free, biologically active leptin. OB-Re influences significantly the leptin kinetic action and consequently its pharmacodynamic action.Material: the source of OB-Re was human serum; the mixture was prepared from 700 healthy volunteers of different age, sex and body weight. Methods: OB-Re was purified from human serum by immunoaffinity chromatography on the column with immobilized mouse monoclonal antibody. OB-Re was eluted from the column by using acid buffer (0.1M NaCl, 0.1M HCl, pH=2), the elution fraction was neutralized by PBS and The purity of the protein was confirmed in SDS PAGE and protein concentration measured by BCA method. We prepared total 5.5 ug of OB-Re, the protein concentration (BCA meth
