We compared the effect of alpha-tocopheryl succinate (TOS) onnsuccinate-dependent respiration in rat liver mitochondria,nhomogenate and permeabilized hepatocytes in both a couplednand uncoupled state. In isolated mitochondria, a significantninhibitory effect was observed at a concentration of 5 ?M, in livernhomogenate at 25 ?M and in permeabilized hepatocytes atn50 ?M. The inhibitory effect of TOS on succinate respiration in annuncoupled state was less pronounced than in a coupled state innall the experimental models tested. When the concentrationndependence of the TOS inhibitory effect was tested, the mostnsensitive in both states were isolated mitochondria; the mostnresistant were permeabilized hepatocytesn
