    Effect of the immobilisation of DNA aptamers on the detection of thrombin by means of surface plasmon resonance

    We report a surface plasmon resonance (SPR) sensor for detection of thrombin via DNA aptamers immobilized on the surface. An effect of the immobilisation method is investigated. Immobilization methods included: (i) chemisorption of aptamers modified withSH groups, (ii) immobilisation of biotin-tagged aptamers via previously immobilized avidin, neutravidin or streptavidin, (iii) aptamer-dendrimer method. Immobilisation of aptamers by the streptavidin-biotin yields the best sensor specificity and sensitivity.

    Senzor s povrchovými plazmony byl použit ke studiu vlivu imobilizační metody DNA aptameru na povrch senzoru pro detekci trombinu. Byly vyšetřovány následující imobilizační postupy: (a) chemisorpce aptameru pomocí SH skupin, (b) vazba biotinovaného aptameru přes avidin, neutravidin a streptavidin, (c) vazba aptameru na dendrimerovou vrstvu. Nejvyšší citlivosti senzoru a specificity bylo dosaženo za použití imobilizačního postupu streptavidin-biotin.
