    Surpassing the Diffraction Limit in Label-Free Optical Microscopy

    This perspective article outlines our vision for future directions in label-free super-resolution microscopy. Drawing insights from the success of fluorescence-based super-resolution techniques, we identify parallels in their label-free counterparts and highlight key challenges in the field. We argue here that the progress in optical instrumentation and advanced microscopy methods achieving single-molecule sensitivity, subdiffractional resolution, and high temporal resolution without labels enables observing processes that are either too complex, too fast, or simply not compatible with the labeling. Leveraging a comprehensive review as a foundation, we extrapolate potential developments and, inspired by our recent work, introduce the concept of iSCAT-PAINT combining interferometric scattering microscopy with the concept of PAINT microscopy. This approach resolves fluctuations of interacting single molecules to reveal subdiffractional details of biological structures
