Effects of selected endocrine disruptors on meiotic maturation, cumulus expansion, syntesis of hyaluronan and progesterone by porcine oocyte-cumulus complexes

  • Original language description

Before ovulation, cumulus cells synthesize a large amount of hyaluronan (HA) that is organized in an extracellular matrix. This process is called cumulus expansion. We assessed effects of endocrine disruptors (bisphenol A, BPA, 4-chloro-3-methyl phenol, CMP, di(2-ethylhexyl) phthalate, DEHP, and benzyl butyl phthalate, BBP) on FSH-induced meiotic maturation of porcine oocyte-cumulus complexes (OCC) cultured in vitro. BPA and CMP, significantly affected meiotic maturation of OCC. The number of oocytes that underwent GVBD; 78.7% and 72.4% as well as the rate of oocytes that reached M II; 50% and 53.6%, were decreased compared to control: GVBD-89.6% ; M II- 81.5%. FSH-stimulated expansion of cumulus was altered by the highest concentration of BPA and CMP (70% and 64% vs. 80.3% in control). Although BPA did not alter FSH-stimulated HA synthesis by cumulus cells, its incorporation within the complex was reduced to a half of control value. Progesterone production by OCC was significantly chan

Vliv vybraných endokrinních disruptorů na meiotické zrání, expanzi kumulu, syntézu hyaluronové kyseliny a progesteronu oocyt-kumulárními komplexy prasat.

  • Czech description

Sledovali jsme vliv endokrinních disruptorů (bisphenol A, BPA; 4-chloro-3-methyl phenol, CMP;di(2-ethylhexyl) phthalate, DEHP; a benzyl butyl phthalate, BBP) na FSH-stimulované meiotické zrání a expanzi oocyt-kumulárních komplexů (OCC) prasat v průběhu in vitro kultivace. BPA i CMP, významně ovlivnily meiotické zrání oocytů.
