Cytosine methylation of plastid genome in higher plants. Fact or artefact?
Original language description
The presence of methylcytosine residues in chloroplast genome has been investigated by restriction enzyme analysis and bisulfite genomic sequencing. We found that only CCWGG sites were partially refractory to the cleavage with EcoRII enzyme. However, bisulfite genomic sequencing failed to show the presence of modified cytosine along 3 region of the rbcL gene. It is likely that the EcoRII refractivity of DNA was caused by specific cleavage properties of this enzyme rather than by methylation of its target site. In contrast to nuclear genome chloroplast genome does not seem to contain significant amounts of methylcytosine residues
