The importance of platinum drugs in cancer chemotherapy is underscored by the clinical success of cisplatin and its analogues. Nevertheless, drug resistance can occur by several ways. This review focuses on mechanisms of resistance and sensitivity of tumors to conventional cisplatin associated with DNA modification. We also discuss molecular mechanisms underlying resistance and sensitivity of tumors to the new platinum compounds synthesized with the goal to overcome resistance of tumors to established platinum drugs.

Czech description
Význam platinových chemoterapeutik v léčbě nádorů je podložen klinickým úspěchem cisplatiny a jejích přímých analog. I přesto se však může vyskytnout k těmto léčivům rezistence, způsobená různými faktory. Tento přehled je zaměřen na mechanismy rezistencea citlivosti nádorů ke klasické cisplatin, související s vazbou na DNA. Diskutujeme také molekulární mechanismus rezistence a citlivosti nádorů vůči novým sloučeninám, které byly připraveny s cílem překonat rezistenci nádorů k dosud používaným platinovým léčivům.
