    Effect of H1-antihistamines on the oxidative burst of rat phagocytes

    Phagocytosis is the essential defensive mechanism against microbial pathogens. This function is ensured by several types of cells with neutrophils in the lead. Phagocyte-derived reactive oxygen species (ROS) belong to the basic microbicidal mechanisms. As observed in our previous study, H1-antagonist dithiaden inhibits the production of ROS by phagocytes. The aim of this study was to compare the inhibition effect of dithiaden with effects of four selected H1-antihistamines of the 2nd generation - loratadine, acrivastine, astemizole and ketotifen fumarate. Acrivastine and loratadine do not suppress the key microbicidal system of phagocytes, ketotifen fumarate inhibited the respiratory burst of phagocytes similarly to dithiaden. Astemizol showed even higher inhibition effect in comparison with dithiaden.

    Fagocytóza je nezbytným mechanismem proti mikrobiálním patogenům. Tato funkce je zajišťována různými typy buněk, zejména neutrofily a jimi produkované reaktivní kyslíkové metabolity (ROS) patří k základním mikrobicidním mechanismům. V předchozí studii jsme zjistili, že H1-antagonista dithiaden inhibuje produkci ROS fagocyty. Cílem této studie bylo srovnat inhibiční účinek dithiadenu se čtyřmi vybranými H1-antihistaminiky 2. generace - loratadine, acrivastinem, astemizolem a ketotifen-fumaratem. Acrivastine a loratadine nepotlačoval mikrobicidní systém fagocytů, ketotifen-fumarate inhiboval oxidativní vzplanutí podobně jako dithiadena a astemizol vykazoval silný inhibiční efekt ve srovnání s dithiadenem.
