Original language description
We showed that frozen/thawed S-phase cells have collapsed replication forks, regardless of the used cryoprotectant. This may have serious implications for assisted reproduction. We also identified changes in higher-order chromatin structure associated with the freeze-thaw process, and correlated them with the viability of frozen and thawed cells. Both in replicating and non-replicating cells, freezing and thawing altered the chromatin structure in a cryoprotectant-dependent manner. Our results suggest that chromatin condensation plays a positive role in cell survival.
