    Changes in extracellular space size and geometry in APP23 transgenic mice - a model of Alzheimer?s disease

    We studied diffusion in the brain of APP23 mice, an animal model of Alzheimer?s disease. We employed two methods: diffusion-weighted MRI and the real-time iontophoretic tetramethylammonium (TMA) method. As revealed by the TMA method, the presence of amyloid plaques in the extracellular space (ECS) was associated with increased ECS volume, more in females than in males. Changes in ECS volume corresponded to an increase in the apparent diffusion coefficient (ADC) of water, as measured by magnetic resonance. In females, there was a significant decrease in the ADC of TMA, reflecting impaired ECS diffusion. The diffusion changes can affect extrasynaptic transmission, neuron-glia interaction and disease progression.

    Studovali jsme difúzi v mozku myší kmene APP23, zvířecího modelu Alzhemerovy nemoci. Použili jsme dvě metody: difúzně váženou magnetickou rezonanci a iontoforetickou metodu sledování difúze tetrametylamonia (TMA) v reálném čase. Pomocí TMA metody jsme zjistili, že přítomnost amyloidových plaků v extracelulárním prostoru (ECP) vede ke snížení objemu ECP, a to více u samiček než u samců. Změny objemu ECP odpovídaly změnám aparentního difúzního koeficientu vody, změřeného magnetickou rezonancí. U samiček došlo v důsledku snížené difuzivity v ECP k poklesu aparentního difúzního koeficientu TMA. Změny difúze mohou ovlivnit extrasynaptický přenos, vzájemnou interakci neuronů a glií a vývoj nemoci.
