    Characterization of human seminal plasma proteins homologous to boar AQN spermadhesins

    Spermadhesins play a role in sperm capacitation and primary contact of sperm and egg. We found human seminal plasma proteins immunobiochemically related to boar AQN and AWN spermadhesins. We further characterized the AQN spermadhesin-related proteins, designated hSA (human spermadhesin-like) proteins. We immunodetected three relative molecular-mass forms of hSA cross-reacting with rabbit antibody against AQN spermadhesins, each comprising three isoelectric isoforms. Gel chromatography of human seminal plasma indicated that hSA proteins formed high molecular-mass complexes with other hSA proteins or with lactoferrin and/or its fragments. Similarity of biochemical properties of hSA proteins and those of boar AQN spermadhesins imply their structural relatedness. However, localization of hSA on the sperm neck and tail suggests their different biological role from that of boar AQN spermadhesins located on the sperm head.

    Spermadhesiny se účastní kapacitace spermie a primárního kontaktu spermie s vajíčkem. Zjistili jsme, že lidská semenná plasma obsahuje proteiny imunobiologicky podobné kančím spermadhesinům AQN a AWN. Blíže jsme charakterizovali lidské proteiny podobné kančím AQN a nazvali je hSA. Na Western blotu jsme detekovali tři hSA proteiny křížově reagující s protilátkami proti kančím AQN spermadhesinům, z nichž každý obsahoval tři izoformy s odlišným pI. Gelová chromatografie lidské semenné plasmy ukázala, že hSA proteiny tvoří vysokomolekulární komplexy mezi sebou nebo s laktoferinem a jeho štěpy. Podobné biochemické vlastnosti hSA proteinů a kančích AQN spermadhesinů naznačují jejich strukturní podobnost. Lokalizace hSA proteinů na bičíku spermie a kančích AQN na hlavičce spermie však napovídají o rozdílné funkci těchto proteinů.
