    Transition to seizure via critical slowing in a chronic model of temporal lobe epilepsy

    Aims: Abrupt changes of the behavior of a complex system, such as onset or offset of an epileptic seizure in the brain, may be preceded by progressive loss of stability which can manifest as a subtle increase in temporal correlation of a state variable of the system. This phenomenon, called critical slowing, was shown to precede offsets of seizures in the EEG of patients with epilepsy. We investigated whether the transition to seizure in a chronic model of epilepsy follows this dynamical path. Methods: Epileptic rats were implanted electrodes into hippocampi and cortices and video-EEG monitored for >2 weeks. From the EEG, we extracted 2-hour pre-seizure epochs and estimated Hilbert envelope. In 10s segments of slow irregular activity, we calculated autocorrelation coefficient (ACC) at 15ms lag. The last 20 minutes of each pre-seizure period were fitted by a straight line. Results: We have analyzed 352 pre-seizure periods. In 4 animals, we found a significant increase of the ACC, in one animal, we found a significant decrease of the ACC and in 4 animals no significant trend was found. Conclusions: We have shown that transition to seizures in vivo can be preceded by critical slowing marking the progressive loss of the resilience of the epileptogenic tissue. Insight into the dynamical principles which govern the seizures genesis has substantial implication for a better understanding of the neural and cellular mechanisms involved seizure in generation.
